Higher Organization of Chromosomes: Fascinating Target for Scientists

DNA which has important roles for maintenance and expression of genetic information, changes its fibrous shape to highly compacted state called a chromosome during cell division for equal distribution of genetic information. Chromosomes were found within a cell in the middle of 19 century. Now DNA and its higher order structure of nucleosome structures within a chromosome are well characterized in atomic resolution, however, chromosome higher order structure itself has not been elucidated yet (Fig. 1). We have tried to elucidate the chromosome higher order structure for the chromosomes isolated and prepared by PA method and Critical Point Drying (CPD) method using Ga FIB/SEM. As a result, we have observed the nucleosome-like signals within a chromosome (Fig. 2). Although it is not achieved to elucidate the whole structure of a chromosome, we believe that the nano- technology is a promising method to elucidate detailed structures of biological samples such as chromosomes.
